退化与正常山核桃林土壤细菌多样性比较研究

Comparison on Soil Bacteria Diversity between Degrated and Healthy Carya cathayensis Forests

  • 摘要: 2014 年10 月,在浙江省临安市选择4 个退化山核桃Carya cathayensis 林样地和2 个正常山核桃林样地,采土样后用16SrDNA 测序技术分析两种林地土壤细菌多样性。结果表明,6 个样地土壤细菌共有36 门75 纲96目224 科924 属。在门的水平上,退化和正常林地细菌主要类群基本一致,主要类群有变形菌门Proteobacteria,酸杆菌门Acidobacteria,拟杆菌门Bacteroidetes,绿弯菌门Chloroflexi,厚壁菌门Firmicutes,疣微菌门Verrucomicrobia,放线菌门Actinobacteria 和浮霉菌门Planctomycetes,但不同林地中各类群相对丰度明显不同;在属的水平上,退化和正常林地差别很大。Alpha 多样性分析显示,6 个样地Shannon 指数,ACE 指数,Chao1指数的排序各不相同,分析说明山核桃林退化程度与土壤细菌数量和多样性无关。相关分析结果表明,ACE 指数与交换性H+显著相关,其余土壤物理化学性质与细菌多样性指数均无显著相关性。

     

    Abstract: Four sample plots in degraded Carya cathayensis stand and 2 in normal ones were established in October 2014 in Lin’an district, Zhejiangprovince. Soil samples were sequenced by 16SrDNA for comparison on bacteria community diversity of different sample plots. The results indicatedthat there were 924 genera of 224 families and 36 phylums, 75 classes, 96 orders in sampled plots. The main phylums were the same in the testedplots, included Proteobacteria, Acidobacteria, Bacteroidetes, Chloroflexi, Firmicutes, Verrucomicrobia, Actinobacteria and Planctomycetesbut withdifferent relative abundance . The distribution of main genera had great difference between degraded and normal plots. The Shannon index, the ACEindex and Chao1 index showed that it had no relationship between degradation of sampled stands with bacteria richness and diversity. Correlationanalysis demonstrated that ACE index had evident relation with H+, the other physiochemical properties of the tested had no significant correlationwith bacteria diversity index.
