5份桉树种质耐寒性指标的测定及其评价

Determination andEvaluation on Cold Resistance of 5 EucalyptusGermplasm

  • 摘要: 以5份桉树种质(邓恩桉Eucalyptus dunnii20784,邓恩桉E.dunnii 99/01,溪谷桉E.badjensis 02/08,邓恩桉E.dunnii 98/27,柳叶桉E.saligna 01/19)的离体叶片为试验材料,通过低温(-7℃)胁迫,测定叶片超氧化物歧化酶(SOD)活性、过氧化物酶(POD)活性、膜脂过氧化产物丙二醛(MDA)含量、可溶性蛋白(SP)含量、半致死温度(LT50),采用相关性分析、主成分分析和聚类分析评价其耐寒性。结果表明:5份桉树种质的低温半致死温度分别为-4.3,-5.31,-3.6,-5.16,-4.73℃。相关性分析显示,SOD活性、POD活性、SP含量均与耐寒性呈正相关,其中SOD活性与耐寒性显著正相关(r=0.982**);对各指标主成分分析表明,SOD活性和POD活性2个主成分解释了原变量85.52%的变异信息,根据综合指数的抗寒性排序为2号邓恩桉E.dunnii99/01>4号邓恩桉E.dunnii 98/27>1号邓恩桉E.dunnii 20784>3号溪谷桉E.badjensis 02/08>5号柳叶桉E.saligna01/19;通过聚类分析,将5份桉树种质抗寒性划分为2类,其中耐寒性较强的种质是2号和4号,耐寒性较弱的是1号、3号和5号。由此得出结论:SOD活性和POD活性是反映5份桉树种质抗寒性的两个重要指标,2号邓恩桉和4号邓恩桉为相对耐寒种质,更适合浙江生长。

     

    Abstract: Seeds of Eucalyptus dunnii20784, E.dunnii99/01, E.badjensis02/08, E.dunnii98/27, E.saligna 01/19 were imported from Australia in July of 2000,sowing and seedling was carried out in Hangzhou of Zhejiang in January of 2013.40 leaves were collectedfrom each germplasm and treated by -7℃ and 4℃in July of 2014 for determination of SOD, POD, MDA, SP and LT50.Correlation analysis on determined index demonstrated that SOD, PODand SP had positive relation with cold resistance, especially SOD had significant relation.Principle component analysis indicated that SOD and POD occupied 85.52% of variable.Cold resistance of tested germplasm was ordered by comprehensive index.Cluster analysis concluded that cold resistance of 5 germplasm could be divided into 2 groups.
