Box-Behnken法优化大叶榉树黄酮类超声辅助提取方法及初步鉴定

Optimization of Ultrasonic Extraction of Flavonoids from ZelkovaschneiderianaLeaves by Box-Behnken

  • 摘要: 为了探讨大叶榉树Zelkovaschneideriana叶片黄酮类成分,以1年生大叶榉树叶片为材料,在超声波辅助下,通过单因素和Box-Behnken试验,探究并优化了大叶榉叶片黄酮类乙醇提取方法;采用常规显色法,对大叶榉树叶片中黄酮类物质种类进行初步鉴定。结果表明:(1)优化后的大叶榉树黄酮类提取方法为:乙醇浓度63%、液料比31:1mL?g-1、提取时间1.7h,大叶榉树黄酮类提取量为(31.067±0.11)mg?g-1;(2)初步鉴定发现:大叶榉树叶片中含有黄酮、具有邻二酚羟基结构的黄酮类化合物、具有邻二苯酚羟基或兼有3-OH,4=O或5-OH,4=O的化合物;可能有黄酮醇、二氢黄酮、二氢黄酮醇等;不含查尔酮和橙酮。

     

    Abstract: On August 26, 2017, collections were implemented on1-3 leaves of 1-yearZelkovaschneiderianain greenhouse for experiment on extraction of flavonoids.Optimization of ultrasonic extraction of flavonoids was designed by box-behnkenwith three factors at three levels.The result demonstrated that the best technology for extraction was as follows, ethanol concentration of 63%, liquidratio of 31:1mL?g-1, extraction time of 1.7 hour, with the yield of flavonoids of (31.067±0.11)mL?g-1.Preliminary identification of extracted flavonoids showed flavones, flavonoids with adjacent two phenolic hydroxyl structures, and compounds with two phenol hydroxyl groups or 3-OH, 4=O or 5-OH, 4=O, and possible flavonoils, dihydroflavones and dihydroflavonols, but no chalcone and aurones.
