11种樟科树枝水提物对樟芝液体培养的比较

Comparison of Aqueous Extracts from 11 LauraceaeBranches on Liquid Culture of Taiwanofungus camphorates

  • 摘要: 通过樟芝Taiwanofungus camphorates菌丝体生物量和总三萜、总多糖含量来分析11种樟科Lauraceae植物的水提物对樟芝液体培养的影响。结果表明,11种樟科植物水提物对樟芝菌丝体的生长均有不同程度的促进作用,其中新木姜子Neolitseaaurata,猴樟Cinnamomumbodinieri,天竺桂C. japonicum和华南桂C. austrosinense的水提物有较为明显的促进作用;11种樟科植物对樟芝菌丝体总三萜含量的影响差异较大,其中新木姜子、兰屿肉桂C. kotoense和猴樟的水提物对总三萜含量的影响较大,分别比对照(基础液体培养基培养的樟芝菌丝体)增加了13倍、10倍和10倍,比前期优化的液体培养分别提高了85.0%,78.5%和78.5%,天竺桂水提物对总三萜含量的影响略微大于前期优化的培养基,而其他的8种樟科植物均有不同程度的促进作用。但这11种樟科植物对樟芝菌丝体总多糖含量的影响均不及前期优化的液体培养基,新木姜子、猴樟的水提物对总多糖含量的影响更为接近前期优化的培养基,但是与对照组相比樟C. camphora和浙江樟C. chekiangense的水提物则表现出一定的抑制作用。通过对樟芝菌丝体的生物量、总三萜和总多糖进行分析,结果显示添加新木姜子水提物对于樟芝的液体培养促进作用最为显著,其次是猴樟。综合分析3个属11种樟科植物的核型、海拔、自然分布等多因素发现,促进作用最为明显的新木姜子与牛樟Cinnamomumkanehirae的核型相同、海拔高且在台湾有自然分布,同时证实了与牛樟不同核型的樟和黄樟C.parthenoxylon的促进作用较小,且促进作用表现出了与海拔成反比,与台湾自然分布或台湾亲缘性成正相关,说明这3个因素均会影响到樟科植物内的成分,推测这些成分中可能具有促进樟科植物的樟芝菌丝体生长等作用。

     

    Abstract: Our research analyzed the effect of aqueous extract of 11 species of Lauraceae plants on Taiwanofungus camphorates liquid culture through itsmycelium biomass and total triterpenoid, total polysaccharide content . The results show that the11 species of Lauraceae plants have different degrees of stimulatory effects on the mycelial growth of T. camphorates. Among them, the aqueous extracts of N. aurata, C. bodinieri, C. pedunculatum and C. austrosinense have relatively obvious stimulatory effects. The effects of aqueous extracts from 11 species of Lauraceae plants on total triterpene content of mycelium of T. camphorates are quite different, among them, N. aurata, C. kotoense and C. bodinieri have obvious stimulatory effects, the total triterpene contentof mycelium of these aqueous water extracts are 13 times, 10 times and 10 times of the control group (the T. camphorates mycelium from base liquid medium) respectively, and the content increase by 85% , 78.5% and 78.5%.The effect of aqueous extractsfrom C. pedunculatum is slightly greater than that of the pre-optimized liquid medium, while the aqueousextracts from other 8 Lauraceae plants have smaller stimulatory effect in different degrees. The effects of aqueous extracts from 11 species of Lauraceae plants on the total polysaccharide content of T. camphorates mycelia are not as good as those of the pre-optimized liquid medium. The effects ofaqueous extracts from N. aurata and C. bodinieri on the total polysaccharide content are relatively closer to the effect of pre-optimized medium. However ,the aqueous extracts from C. camphora and C. chekiangense show a certain inhibitions when compared with the control group. Through the analysis of the biomass, total triterpenes and polysaccharides of the mycelium of T. camphorates, the results show that the addition of aqueous extract from N. auratahave the most obvious stimulatoryeffect on the liquid culture of T. camphorates, followed by C. bodinieri. Comprehensive analysis of plants karyotype, elevation and natural distribution of the 11 species of Lauraceae, revealsthat N. aurata, which havethe most obvious stimulatory effect , present the same karyotype , high altitude and natural distribution in Taiwan as that of C.kanehirae.. At the same time, it has been proved that C. camphora and C. parthenoxylon have less promoting effect cause its karyotype are different from C. kanehirae, and the promoting effect is in inverse proportion to the altitude, in direct proportion to the natural distribution and relatedness of Taiwan. It is suggested that the three factors would affect the composition of Lauraceae plants,and further speculated that these components may promote the mycelium growth of Lauraceae plants.
