外源激素对银杏雌株开花性状和开花相关基因表达的影响

Effects of Exogenous Hormones on Female Ginkgo biloba’s Flowering and Expression of Flowering Related Genes

  • 摘要: 为探讨植物外源激素对银杏Ginkgo biloba 雌株结实和开花的影响,揭示外源激素如何通过影响开花相关基因的表达进而调控银杏花芽分化的分子机理,对银杏花芽分化诱导期的成年雌株进行GA3(赤霉素)和PP333(多效唑)2 种外源激素喷施实验,并利用RT-qPCR 手段分析银杏开花相关基因在不同外源激素处理间转录水平的差异,分析基因相对表达量与结果枝花芽、叶芽数的相关性。结果表明,在银杏花芽分化诱导期多次喷施200 mg·L-1的GA3 能在一定程度上抑制雌株开花;GA3 和PP333 两种激素处理对银杏雌株混合芽GinNdly 基因的表达没有明显的诱导作用,但对GinLFY 和GBM5 基因的表达具有显著的诱导上调;相关性分析结果表明,GinNdly 基因的表达与每米结果枝花芽数呈负相关,GinLFY 和GBM5 基因的表达具有较高的同步性,且与每米结果枝花芽数呈正相关。据此推断,GinLFY 基因与银杏雌株的开花密切相关且对外源激素的响应较为敏感,上游开花相关基因GinLFY 的表达调控GBM5 基因的相对表达,从而影响银杏的开花结实。本研究为揭示外源激素通过影响开花相关基因的表达进而调控银杏花芽分化的分子机理研究提供了证据。

     

    Abstract: In April 2018, 15 female bearing trees of Ginkgo biloba with 10-year age were selected for experiment. They were applied organic fertilizer once respectively during period of dormancy and flowering each year. Meantime, bearing branches were cut for flower and leaf bud number. In June 2018, these trees were sprayed 3 times of 200 mg/L GA3, 100 mg/L PP333 and water for control with interval of 10 days. In April 2019, bearing branches were obtained for extraction of RNA and test of RT-qPCR. The results showed that repeated application of 200 mg·L-1 GA3 during the differentiation induction period of G. biloba could inhibit the flowering. GA3 and PP333 treatments had no induction effect on the expression of GinNdly gene in the female mixed bud, but they had significant induction effect on the expression of GinLFY and GBM5 genes. Among them, the GinLFY gene expression after GA3 and PP333 treatment increased by 7.6 and 5.4 time, and the GBM5 increased by 904 and 551 time, respectively. Correlation analysis showed that the expression of GinNdly gene was negatively correlated with average number of flower buds per meter (ANFB), while the GinLFY and GBM5 genes was positively correlated with ANFB. Therefore, GinLFY gene was closely related to flowering and sensitive to the response of exogenous hormones, which was likely to be the peripheral factor regulating the flowering and fruiting of G. biloba. The expression of GinLFY, an upstream flowering related gene, regulated the relative expression of GBM5 gene, thus affecting the flowering and fruiting of G. biloba.
