山杏叶中角鲨烯提取工艺的研究

Extraction Technology of Squalene from Armeniaca sibirica Leaves

  • 摘要: 以山杏Armeniaca sibirica 叶为原料,采取SEF-CO2(CO2 超临界萃取法)法提取山杏叶油,用气相色谱-质谱(GC-MS)对其化学成分进行分析。共鉴定出20 种化合物,包括角鲨烯、烷烃、不饱和脂肪酸、酯类、植醇等化合物,其中角鲨烯占精油总组分的38.49%,可见山杏叶油中以角鲨烯为主要成分。以单因素试验、正交试验对山杏叶中角鲨烯提取工艺进行研究,确定提取山杏叶角鲨烯的最佳工艺条件为:提取溶剂为石油醚60 ~ 90℃,提取温度为60℃,料液比为1:10,提取时间为2.5 h。在最佳工艺条件下从山杏叶中提取出的角鲨烯含量最高可达2.19 mg·g-1。

     

    Abstract: In October 2016, leaves were collected from 20-year Armeniaca sibirica in Chengde, Hebei province. Leaf oil was extracted by supercritical CO2 extraction technology, and analyzed by gas chromatography-mass spectrometry. 20 compounds were identified, including squalene, alkane, unsaturated fatty acids, esters, phytol, etc. Among them, squalene occupied 38.49% of essential leaf oil. Extraction technology of squalen was conducted by single factor and orthogonal design tests. The result demonstrated that the best technology was as follows: extraction solvent of petroleum ether (60-90℃), extraction temperature of 60℃, solid-liquid ratio of 1:10, and extraction duration of 2.5 hour, squalene content extraedted of 0.219%.
