野生灵芝的分离鉴定及林下栽培技术研究

Isolation and Identification of Wild Ganoderma lucidum and Cultivation Under Forest

  • 摘要: 灵芝Ganoderma lucidum 是我国重要的珍贵食药用菌。本研究从浙江省龙泉市森林中采集得到8 个野生灵 芝子实体,通过菌丝体分离,获得了相应8 个灵芝菌种,比较不同菌株在PDA 培养基和原种培养基的生长速度, 发现采自板栗Castanea mollissima 上的菌株LQ06 的生长最快。对其进行形态学和ITS 序列比对分析,明确了菌 种LQ06 为灵芝属灵芝种真菌G. lucidum Dai 2272。比较了灵芝LQ06 林下栽培过程中不同段木栽培基质、不同林 地郁闭度及塑料棚设置等因素对灵芝生长及产量的影响。结果显示,以麻栎Quercus acutissima 为段木栽培基质进 行栽培,所得灵芝的生长性状及孢子粉产量显著高于以枫香树Liquidambar formosana、板栗和青冈Cyclobalanopsis glauca 为段木栽培基质的(P<0.05);在林分郁闭度为0.6 和0.8 的条件下,灵芝的生长性状及孢子粉产量均优 于郁闭度为0.4 的条件;设置灵芝棚可显著促进灵芝生长和提高孢子粉产量,林下栽培得到的灵芝菌柄平均长度 为12.04 cm,菌盖平均直径为17.92 cm,生物转化率为5.63%,每个成熟灵芝子实体平均干质量为163.43 g,孢子 粉平均质量为152.37 g,粉/芝比为93.23%。研究结果对灵芝野生资源开发及山区林下经济发展具有理论和应用价值。

     

    Abstract: In August 2017, 8 fruiting bodies of wild Ganoderma lucidum were collected in Longquan, Zhejiang province and 8 strains were isolated. Experiments were conducted on cultivation of isolated stains on PDA and primary stock substrate for comparing their growth. The result showed that stain of LQ06 collected from Castanea mollissima had the fastest growth. Analysis on its morphology and sequences of the internal transcribed spacer (ITS) region demonstrated that LQ06 was G. lucidum Dai 2272. Cultivation tests of LQ06 were carried out in Longquan in 2018 and 2019, with different cut logs, under different canopy densities and green house. The result concluded that LQ06 cultivated on cut log of Quercus acutissima had the best growth and highest spores yield, with crown densities of 0.6 and 0.8. Cultivation in greenhouse could significantly increase the growth and spores yield. The average length of stipes and diameter of caps was 12.04 cm and 17.92 cm, with 5.63% of biological efficiency. The average weight of each dry fruiting body and spore yield was 163.43 g and 152.37 g, resulting in the ratio of 93.23%.
