AB-8树脂纯化竹叶黄酮工艺及其抗氧化活性研究

Experiment on Purification Technology of Bamboo Leaf Flavonoid by AB-8 and Its Antioxidant Activity

  • 摘要: 采用AB-8 大孔树脂对竹叶黄酮提取物进行纯化,考察其动态吸附-解吸动力学特性,并对AB-8 大孔树脂 动态柱层析的工艺条件进行优化;以上样吸附率和洗脱解吸率为检测指标,通过动态吸附实验考察上样液质量浓 度、上样流速和上样体积、洗脱液体积分数、洗脱体积和洗脱流速来优选最佳工艺条件;采用NaNO2-Al(NO3)3 比色法测定总黄酮含量,检测波长为510 nm,通过DPPH 自由基清除率和·OH 清除率来评价竹叶黄酮纯化前后的 抗氧化活性。结果表明,动态柱层析纯化的最佳工艺条件为:上样液质量浓度为1.2 mg·mL-1、上样体积为60 mL、 上样流速2 BV·h-1(BV:柱体积);乙醇洗脱剂体积分数为70%、洗脱体积为60 mL、洗脱流速为3 BV·h-1。在 此工艺条件下对竹叶黄酮进行纯化,竹叶黄酮纯度由28.39%提高至77.42%。抗氧化活性研究结果表明,竹叶黄 酮对DPPH 自由基以及·OH 都具有较好的清除效果,清除率均随着竹叶黄酮样品质量浓度的增加而增强;纯化后 的竹叶黄酮在浓度为1.0 mg·mL-1 时,对DPPH 自由基的清除率最高,为96.68%,对·OH 的清除率也达到最高, 为29.41%。用最佳工艺条件纯化竹叶黄酮粗提物,简单便捷,具有可行性,体外抗氧化活性效果明显,可为开发 竹叶黄酮美白,抗衰老等产品提供参考。

     

    Abstract: AB-8 macroporous resin was used to purify bamboo leaf flavonoid (BLF). With adsorption rate and desorption rate of BLF as the indicators, experiments were conducted on the optimal technology for selection of the mass concentration, the flow rate and volume of loading solution, the ethanol concentration, the flow rate and volume of elution. The result demonstrated that BLF increased from 28.39% to 77.42% under the technology as follows: the concentration of loading solution was 1.2 mg/mL, the loading volume was 60 mL, and the flow rate was 2 BV/h, the ethanol concentration was 70%, the volume of eluent was 60 mL, and the flow rate was 3 BV/h. The antioxidant activity showed that BLF had the highest scavenging rate of 96.68% for DPPH and 29.41% for ·OH.
