3种花色类型滇黄精的遗传关系研究

Genetic Relationship of Polygonatum kingianum with Three Different Flower Colors

  • 摘要: 滇黄精Polygonatum kingianum 是我国传统名贵中药材,也是最具云南特色的药用植物资源。在大理州大理 市挖色镇进行滇黄精资源收集和扩繁过程中,发现滇黄精资源在花被颜色上存在紫红花、白花和白花带紫斑3 种 类型,其中花被白花带紫斑为新发现类型。为明确3 种花色滇黄精的遗传关系,对3 种花色滇黄精的48 个植株进 行ISSR 分子标记分析。结果表明,11 条ISSR 引物对48 个植株样品进行扩增,共获得103 条清晰条带,其中多 态性条带有99 条,多态性比率为96.1%,遗传一致度为0.89 ~ 0.91。遗传差异和UPGMA 聚类分析显示,白花类 型(P1)、白花带紫斑类型(P2)与紫红花类型(P3)的滇黄精均各自独立聚为一组,遗传距离上白花带紫斑类 型(P2)与紫红花类型(P3)的亲缘关系较近,可知白花带紫斑类型(P2)是紫红花类型(P3)和白花类型(P1) 的过渡类型或是白花类型与紫红花类型自然授粉所产生子代的一种杂合状态。本研究结果为滇黄精资源的遗传关 系研究及分类鉴别奠定了一定基础。

     

    Abstract: In July 2019, Polygonatum kingianum with flower of purple safflower, white flowers and purple spots on white flowers were collected 48 individuals each in Dali, Yunnan province for ISSR molecular marker analysis. The results showed that a total of 103 bands were obtained from 48 P. kingianum individuals amplified with 11 ISSR primers, among which, 99 bands were polymorphic, and the genetic identity was 0.89-0.91. The genetic difference and UPGMA clustering analysis showed that the white flower genotype (P1), the white flower with purple spot genotype (P2) and the purple safflower genotype (P3) could be grouped into one group, among which, there was closer genetic relationship between P2 and P3.Therefore, the P2 might be the transitional genotype between P3 and P1, or a heterozygous state of the offspring produced by the plants of the white flower genotypes after receiving the pollen of the purple safflower.
