三叶青叶不同萃取物的抗氧化和抗炎活性评价

Experiment on Antioxidant and Anti-inflammatory Activity of Different Extracts from Leaves of Tetrastigma hemsleyanum

  • 摘要: 为研究三叶青Tetrastigmahemsleyanum叶不同萃取物的抗氧化和抗炎活性,并探究其物质基础,本研究利用石油醚、乙酸乙酯、水饱和正丁醇、水萃取三叶青叶乙醇提取物得到4种萃取物,分别测定4种萃取物中的总黄酮、总多酚含量;采用DPPH法、FRAP法和总抗氧化活性法评价三叶青叶不同萃取物的抗氧化活性,在脂多糖诱导的RAW264.7细胞炎症模型上,对该细胞炎症因子NO进行了测定,评价三叶青叶不同萃取物的抗炎活性,并利用高效液相色谱分析相关性。结果表明,三叶青叶乙醇提取物乙酸乙酯萃取物中的总黄酮、多酚含量最高,分别为145.82±3.09mg·g-1、27.18±1.01mg·g-1,具有最强的抗氧化能力,石油醚萃取物具有最强的抗炎能力;相关性分析表明,萃取物中的总黄酮、总多酚含量对其抗氧化能力有显著影响(P<0.01),对抗炎能力无明显影响(P>0.05);HPLC分析表明,芦丁、绿原酸等可能是三叶青叶抗氧化相关成分。以上研究结果定位了三叶青叶提取物抗氧化、抗炎能力突出的萃取物,可为后续三叶青叶抗氧化、抗炎产品的开发利用提供参考。

     

    Abstract: In May 2022, leaves of 3-year Tetrastigma hemsleyanum were collected in Suichang, Zhejiang province. Four extracts were obtained from leaves by different reagents (petroleum ether, ethyl acetate, water saturated n-butanol, water). Determinations were implemented on total flavonoids and polyphenols content, and DPPH, FRAP and total antioxidant activity methods were used to evaluate antioxidant activities of different extracts. The anti-inflammatory activity of the different extracts was evaluated by an LPS-induced inflammation model of RAW 264.7 cells by measuring cytokine nitric oxide. The correlation was further analyzed by HPLC. The result demonstrated that extract by ethyl acetate had the highest content of total flavonoids and polyphenols, 145.82±3.09 mg/g and 27.18±1.01 mg/g, and the strongest antioxidant capacity. Extract by petroleum ether had the strongest anti-inflamatory capacity. Correlation analysis showed that the content of total flavones and polyphenols had evident effect on antioxidant capacity (P < 0.01), but no significant effect on anti-inflammatory capacity (P > 0.05). HPLC analysis resulted that rutin and chlorogenic acid may be antioxidant-related components of extracts from leaves of T. hemsleyanum.
