甲维盐注干施药对马尾松内生真菌和细菌多样性的影响

Effect of Trunk Injection of Emamectin Benzoate on Diversity of Endophytic Bacteria and Fungi in Pinus massoniana

  • 摘要: 以未施药马尾松Pinusmassoniana和甲维盐注干施药5个月后的马尾松主干内生真菌、细菌为研究对象,采用高通量测序技术IlluminaNovaSeq6000对内生细菌(16SrRNAV3+V4)和真菌(ITS)的群落结构和多样性进行分析。结果表明,高通量测序后共得到运算分类单元(operationaltaxonomicunits,OTUs)细菌1169个、真菌7250个,聚类得到细菌27门340属、真菌15门594属;与未施药马尾松相比,施药马尾松共有OTUs数减少71.84%;在门、属水平上内生菌群落结构差异不明显,门水平上,相对丰度最高的细菌为蓝藻门Cyanobacteria和变形菌门Proteobacteria,真菌为子囊菌门Ascomycota、担子菌门Basidiomycota和被孢霉门Mortierellomycota,属水平上,相对丰度最高的细菌为蓝细菌目未分类属unclassified_Cyanobacteriales;Beta多样性分析表明,组间无明显差异(P>0.05);PCoA分析显示,未施药和施药两组处理细菌群落结构差异不显著,真菌群落结构差异显著;Metastats分析表明,注干施药松树中色二孢属Diplodia的相对丰度显著降低(P<0.01)。以上研究结果表明,甲维盐注干施药对马尾松内生真菌和细菌多样性有一定影响,明显降低了色二孢属的相对丰度,有利于马尾松健康,甲维盐注干使用对马尾松安全。

     

    Abstract: On April 20th, 2022, sample plot with 0.33ha of coniferous and broad-leaved mixed forest was selected, and 20 Pinus massonina trees with 20-35-year age were selected for trunk injection of 2.3% micro-emulsion of emamectin benzoate. On September 20th, 2022, bits of 3 injected trees and 3 controls (non-injection) were sampled from trunk and branch. Community structure and diversity of endophytic bacteria (16S rRNA V3+V4) and fungi (ITS) was carried out by high-throughput sequencing technology Illumina NovaSeq 6 000. A total of 1,169 bacterial and 7 250 fungal operational taxonomic units (OTUs) were obtained, with 27 phyla and 340 genera of bacteria, 15 phyla and 594 genera of fungi. The number of OTUs in the injected trees reduced by 71.84% compared with that in the control. The differences in the community structure of endophytic bacteria at the phylum and genus levels were not obvious.
