盐胁迫对12个紫薇品种种子萌芽和幼苗生理的影响

Effect of Salt Stress on Seed Germination and Seedling Physiological Characteristics of Lagerstroemia indica Cultivars

  • 摘要: 为探究盐胁迫对紫薇(Lagerstroemia indica)种子萌芽、幼苗生理的影响,本研究以12个紫薇品种的种子和幼苗为试材,利用NaCl溶液模拟盐胁迫,萌芽期设置0、50、100 mmol·L−1盐浓度处理,幼苗期经筛选后设置0、75、150 mmol·L−1盐浓度处理,测定种子萌发和幼苗生理有关指标,采用隶属函数法和聚类分析对萌芽期和幼苗期的耐盐性进行综合评价。结果表明:随着NaCl浓度的增加,各品种的发芽率(GR)、发芽指数(GI)、活力指数(VI)和相对含水量(RWC)大部分呈下降趋势,其中在高盐胁迫下,‘幻粉’ 的发芽率和发芽指数最高,‘红火球’ 的活力指数最高,‘大花粉晶’ 的相对含水量最高;总叶绿素(ChI)含量和丙二醛(MDA)含量总体呈上升趋势,对 ‘大花粉晶’ 的细胞膜透性受盐胁迫的影响最大;各品种根芽比(R/Sr)和过氧化氢酶(CAT)活性在胁迫时变化复杂,‘红火球’ ‘白雪’ 和 ‘建明红’ 的根芽比呈上升趋势,‘幻粉’ ‘夏日舞娘’和 ‘PETIT PINK’ 的过氧化氢酶活性呈持续上升。综合两个时期各指标进行隶属函数分析,对12个紫薇品种耐盐能力由强至弱排序前三名的依次为 ‘红火球’ ‘幻粉’ ‘紫绶’,排后三名的为 ‘PETIT PINK’ ‘PKES’ ‘大花粉晶’。本研究通过对种质和幼苗材料的初筛与鉴别,发现 ‘红火球’ ‘幻粉’ ‘紫绶’ 三个品种表现出较好的耐盐性,对未来栽植扦插苗进行长期观测并从基因水平探究其耐盐能力提供了理论依据。

     

    Abstract: To explore the effects of salt stress on the seed germination and seedling physiological characteristics of Lagerstroemia indica, seeds and seedlings of 12 cultivars were used as experimental materials. NaCl solutions were applied to simulate salt stress, with concentrations of 0, 50 and 100 mmol·L−1 during the germination stage, and 0, 75 and 150 mmol·L−1 during the seedling stage after preliminary screening. Relevant indicators of seed germination and seedling physiology were measured, and the membership function method and cluster analysis were employed to comprehensively evaluate salt tolerance at both stages. The results showed that the germination rate (GR), germination index (GI), vigor index (VI), and relative water content (RWC) of most cultivars generally decreased with the increasing NaCl concentration. Under high salt stress, ‘Huan Fen’ exhibited the highest GR and GI, ‘Honghuoqiu’ had the highest VI, and ‘Dahua Fenjing’ showed the highest RWC. Total chlorophyll (Chl) content and malondialdehyde (MDA) content generally increased, with the cell membrane permeability of ‘Dahua Fenjing’ being affected most by salt stress. The root-shoot ratio (R/Sr) and catalase (CAT) activity varied complexly under salt stress; the R/Sr of ‘Honghuoqiu’, ‘Baixue’, and ‘Jianming Hong’ increased, while CAT activity in ‘Huan Fen’, ‘Summer Dance’ and ‘PETIT PINK’ continued to rise. Through the membership function analysis of all indicators in the two periods, the salt tolerance of 12 cultivars from strongest to weakest was ranked, with the top three being ‘Honghuoqiu’, ‘Huan Fen’ and ‘Zishou’, and the last three being ‘PETIT PINK’, ‘PKES’ and ‘Dahua Fenjing’. Through the initial screening and identification of germplasm and seedling materials, the cultivars ‘Honghuoqiu’, ‘Huan Fen’ and ‘Zishou’ demonstrated better salt tolerance, providing a theoretical basis for the long-term observation of future planted cutting seedlings and the exploration of their salt tolerance ability at the genetic level.
