响应面法优化内生真菌发酵产紫杉醇工艺

Optimization of Fermentation Conditions for Taxol Production from Endophytic Fungus by Response Surface Methodology

  • 摘要: 选择产紫杉醇的内生真菌EF-04和葡萄糖、豆饼粉、乙酸钠三种营养因子,在单因素试验的基础上,通过Box-Benhnken组合设计和响应曲面分析法优化内生真菌液体发酵产紫杉醇的发酵工艺条件进行紫杉醇人工生产试验。响应面分析结果表明,内生真菌液体发酵产紫杉醇的最佳培养条件为葡萄糖25.54g/L、豆饼粉7.94 g/L、乙酸钠5.64 g/L;此条件下的验证试验表明,实际紫杉醇得率为313.52 μg/L,与理论值基本吻合。

     

    Abstract: Based on the results of single factor experiment, three levels and three factors of Box-Behnken design was employed combining with response surface methodology (RSM) to optimize the fermentation conditions for taxol production from endophytic fungus. The result showed that the optimal fermentation conditions were as follows: glucose 25.54 g/L, soybean powder7.94 g/L and sodium acetate 5.64g/L. Verification test demonstrated that the average yield of taxol was 313.52 μg/L, similar to theoretical value of 331.6 2μg/L.
