正交试验优选白及组培苗生根培养基

Orthogonal Experiment on Medium for Tissue Culture Bletilla striata Seedling Rooting

  • 摘要: 为优化白及(Bletilla striata)组培苗生根技术体系,试验以1/2MS为基础培养基,采用4因素3水平(L9(34))3次重复正交试验,研究NAA、GA3、6-BA及香蕉泥4因素不同组合对白及组培苗生根效果的影响。结果表明,4个试验因素对白及生根的影响大小程度依次为NAA > 香蕉泥 > GA3 > 6-BA,其中NAA、GA3及香蕉泥有促进白及根系生长的作用,6-BA则对白及生根有抑制作用;白及生根培养基的最适配方为:1/2MS+ NAA0.2 mg/L + GA30.5mg/L+香蕉泥30g/L;白及根系生长曲线,总体呈慢—快—慢的“S”型增长。

     

    Abstract: Orthogonal experiments were conducted on effect of different concentration and combination of NAA, GA3, 6-BA and mashed banana in 1/2MS on rooting of tissue culture Bletilla striata seedling. The results showed that NAA, mashed banana and GA3 could promote growth of root system, while 6-BA had inhibitory effect on rooting. The optimal medium for rooting was 1/2MS + 0.2 mg/L NAA + 0.5mg/L GA3 + 30g/L mashed banana. Root growth curve of tissue culture Bletilla Striata seedling was "S" type.
