蒸汽爆破预处理对樟芝液体培养的影响

Effect of Steam Explosion of Cinnamomum bodinieri Branch on Liquid Culture of Ganoderma camphoratum

  • 摘要: 采用 7个维压时间的蒸汽爆破对猴樟 Cinnamomum bodinieri树枝进行预处理,并通过扫描电镜、木质素、纤维素、碳含量和有机氮含量等指标进行分析,再将这 8组猴樟水提物添加到樟芝 Ganoderma camphoratum液体培养基中,研究其对液体培养樟芝菌丝体总三萜、总多糖含量的影响。结果表明,通过蒸汽爆破的预处理,猴樟树枝的形态发生了明显的变化,扫描电镜图显示随着维压时间的增加,大的碎块断裂,逐渐呈现出许多小孔,且纤维素、木质素、碳和氮含量均呈缓慢上升趋势,碳氮比却显著下降并在 300 s时降到真菌培养基的理想水平 25;同时证实蒸汽爆破预处理均可以促进樟芝液体培养产三萜和多糖,最高值比对照组分别提高了 16.80 %和 25.00%,其中当维压时间在 300 s时,总三萜的含量达到最大。研究结果表明汽爆可破坏猴樟的表皮、纤维层,有效提高其有效成分的提取,预示此技术在有效成分提取中具有广阔的发展前景。

     

    Abstract: Experiments were conducted on different duration of steam explosion of branch fragments of Cinnamomum bodinieri for its water extract. Analysis were made on pictures of scanning electron microscope, as well as on lignin, cellulose, carbon content and organic nitrogen content of different treated fragments and control (no explosion).Water extract from different treated fragmentes was added to the liquid culture media for studying the total triterpene and total polysaccharide content of mycelium in liquid culturing.The results show that the form of the fragmentes changed obviously after steam explosion.Scanning electron microscope showed that larger fragments fractured and gradually had many small holes.The contents of cellulose, lignin, carbon and nitrogen increased slowly with longer duration of explosion.But carbon-nitrogen ratio decreased significantly even to the ideal level for fungus culture medium at 300 s.The experiment resulted that steam explosion could promote the mycelium of total triterpenes and total polysaccharide production, the highest had 16.80% and 25.00% more than the control.
