Identification of Cultivars of Torreya grandis Cultivars (Strains) by Multiple Fluorescent SSR Markers

  • In November 2020, fresh tender leaves were collected from 6 cultivars and 3 strains (3 trees each) of Torreya grandis in Pan’an county, Zhejiang province for identification experiment. SSR markers were screened according to the previously established transcriptome database of a T. grandis ‘Xifei’ in order to obtain polymorphic SSR markers. These polymorphic SSR markers were further used for detect genotypic difference among 9 materials. The multiplex PCR assays were further established for the joint use of these polymorphic SSR markers for identifying cultivars and strains based on different genotype combinations. The results showed that 4 genotypes combinations including 289/295 plus 192/222, 277/295 plus 192/222, 277/289/295 plus 192/216/222, and 277/295 plus 192/216 were detected from two polymorphic SSR loci of Tg_U7 and Tg_U734 for 9 materials. The different genotype combinations allowed to identify simultaneously 2 cultivars and 1 strain of T. grandis, namely ‘Yushanyufei’, ‘Pandafei’ and Dadingxiang strain.
