Study on Tissue Culture and Rapid Propagation Technology of Color-leafed Cinnamomum chekiangense

Abstract
With the current-year semi-lignified stem segments with bud points of color-leafed Cinnamomum chekiangense as explants in this study, the sterilization effect on explants in different disinfection times was compared, and how the basic medium types and the proportion of plant-growth regulators impact on the dedifferentiation and redifferentiation of the explants was illustrated.The results are as follows, the suitable disinfection procedure for color-leafed C. chekiangense explants was 75% ethanol for disinfection for 30 seconds + 0.1% HgCl2 for disinfection for 8 minutes, with a survival rate of 85.00% and a infection rate of 13.33%. The basic culture medium suitable for explants lateral bud induction was MS, and when the growth regulator with the ratio of 0.9 mg·L−1 6-BA + 0.5 mg·L−1 IBA was added, the induction time of lateral buds was short, the induction rate reaching 86.67%. The ratio of growth regulators suitable for subculture proliferation was 0.5 mg·L−1 6-BA + 0.05 mg·L−1 NAA +0.3 mg·L−1 GA3, with a proliferation coefficient of up to 3.15 and an average seedling height of 1.32 cm. The IBA concentration suitable for rooting culture was 0.5 mg·L−1, with a rooting rate of 83.33%.The efficiency of tissue culture and rapid propagation for color-leafed C. chekiangense is significantly improved in this experiment, which is beneficial for industrial cultivation of superior color-leafed varieties of C. chekiangense.
